A Myosin-like Protein in the Cortical Layer of the Sea Urchin Egg

چکیده
An interaction between actinand myosin-like proteins and its implication in cellular motilities have been suggested in a variety of cells other than muscle (reviewed by Taylor, 1972, and by Rebhun, 1972) . Actually, those proteins have been extracted and purified from certain kinds of cells . They may be located mainly in the cell cortex (Ishikawa et al ., 1969) . In sea urchin eggs, although an actin-like protein which could interact with rabbit skeletal muscle myosin in a manner similar to muscle actin was extracted and purified from acetone powder of whole cell (Miki-Noumura and Oosawa, 1969), an actomyosin-like component has not been detected in the isolated cortex (Mabuchi and Sakai, 1972) . However, extensive studies on this line have confirmed that a myosin-like component was contained in the 0 .6 M KCl-extract of the isolated cortex of the sea urchin egg . It will be called "ovomyosin" in this paper .

متن کاملCharacterization of sea urchin egg endoplasmic reticulum in cortical preparations.
The cortical endoplasmic reticulum (ER) of sea urchin eggs was localized on isolated egg cortices by staining with aqueous suspensions of the dicarbocyanine "DiI." Immunofluorescence localization of a calsequestrin-like protein was essentially identical; this is consistent with a role for the ER in calcium regulation. The ER often encircles cortical granules, making it well-suited for initiatin...
متن کاملDirect Isolation of the Hyaline Layer Protein Released from the Cortical Granules of the Sea Urchin Egg at Fertilization
Treatment of the eggs of the sea urchin with a 1 M solution of glycerol at fertilization allows the recovery from this solution of the protein released from the cortical granules, including that which would normally give rise to the hyaline layer. The calcium-gelable protein previously extracted from whole eggs and from isolated cortical material was found to be present in the glycerol solution...
متن کاملLow ionic strength solubility of myosin in sea urchin egg extracts is mediated by a myosin-binding protein
We identify a novel myosin-binding protein, designated 53K, which appears to mediate the low ionic strength solubility of myosin in extracts of unfertilized sea urchin eggs. The protein possesses a subunit molecular mass on SDS-PAGE of 53 kD, an S value of 7, may be organized into disulfide-linked oligomers, and is associated with myosin in egg extracts. Both myosin and 53K co-precipitate from ...
متن کاملPhosphoprotein inhibition of calcium-stimulated exocytosis in sea urchin eggs
We have investigated the role of protein phosphorylation in the control of exocytosis in sea urchin eggs by treating eggs with a thio-analogue of ATP. ATP gamma S (adenosine 5'-O-3-thiotriphosphate) is a compound which can be used as a phosphoryl donor by protein kinases, leading to irreversible protein thiophosphorylation (Gratecos, D., and E.H. Fischer. 1974. Biochem. Biophys. Res. Commun. 58...
